Dr. YANG Yungui is a professor at Beijing Institute of Genomics, Chinese Academy of Sciences (CAS). Dr YANG received his BS degree in Microbiology from Fudan University in 1995 and PhD degree in Biochemistry and Molecular Biology from Shanghai Institute of Materia Medica/Shanghai Research Center of Biotechnology, CAS in 2000 with Dr. YANG Shengli . He performed his postdoctoral research in the laboratories of Dr WANG Zhaoqi (International Agency for Research on Cancer-IARC, 2000-2003) and Dr Tomas Lindahl (Nobel Laureate in Chemistry 2015; Clare Hall Laboratories, Cancer Research UK London Research Institute, 2005-2008). He had served as a staff scientist at IARC from 2003-2005. His research interest is to dissect the roles and biological significance of epitranscriptomic RNA modifications with focus on dynamic RNA methylations (i.e. N6-methyladenosine-m6A, 5-methylcytosine-m5C). He has totally published 36 corresponding- and 8 first-author papers in the peer-reviewed journals including Nature, Cell, Nature Cell Biol, Cell Stem Cell, Mol Cell, Cell Res, etc. He was elected as Member of the Academia Europaea in 2020
Introduction
Our research interests mainly focus on epitranscriptomic regulations and mechanisms of RNA methylation. We have contributed in a breakthrough point of expanding new research frontiers of RNA methylation-mediated epitranscriptomics by identifying the mammalian RNA N6-Methyladenosine (m6A) methyltransferase complex METTL3/METTL14/WTAP (Ping et al. Cell Research 2014), selective mechanism of m6A site (Chen et al. Cell Stem Cell 2015), m6A demethylases FTO (Jia et al. Nature Chemical Biology 2011) and ALKBH5 (Zheng et al. Molecular Cell 2013) and regulatory roles of m6A in mRNA splicing, translation and transcription (Zhao et al. Cell Research 2014; Xiao et al. Molecular Cell 2016; Li et al. Cell Research 2017; Yang et al. Cell Research 2018; Yang et al. Cell Research 2019). Our recent works further revealed that m6A modulates spermatogenesis, haematopoietic stem/progenitor cell specification, brain development and even cancer (Zhang et al. Nature 2017; Xu et al. Cell Research 2017; Zhang et al. Cell Research 2018; Jia et al. Molecular Cancer 2019; Wu et al. Cell Research 2019). We further involved in the identification of entacapone as a chemical inhibitor of FTO mediating metabolic regulation (Peng et al. Science Translational Medicine 2019). In addition, we uncovered the regulatory role of m5C in mediating mRNA export via its reader protein ALYREF (Yang et al. Cell Research 2017), and mRNA stabilization via its reader Ybx1 then regulating zebrafish maternal-to-zygotic transition (Yang et a. Molecular Cell 2019) and pathogenesis of bladder cancer (Chen et al. Nature Cell Biology 2019). We further mapped dynamic methylome of internal mRNA N7-methylguanosine (Malbec et al. Cell Research 2019). The discovery of the RNA methylation methyltransferases and demethylases demonstrate that the dynamic and reversible chemical modification on RNA is now considered a novel epitranscriptomic marker of profound biological significance.
